Firstly, I’d like to state that both these workshops will be using the Cytek Aurora platform using Spectroflo software. There are other spectral instruments available from Sony and BD Biosciences but we have the Aurora instruments in our lab. It may seem daunting to switch from conventional cytometry to full spectral cytometry but, the aurora platform is a fairly intuitive and easy to learn. But, more than ever, we are dependent on having good quality controls. It is essential to have completely unstained cells and single colour reference controls. Workshop 2 Panels exploring non-lymphoid tissues present several additional challenges. There may be various populations of cells with different levels of autofluorescence, large amounts of debris and many dead cells. We will discuss: 1. Reference controls 2. Autofluorescence 3. Staining considerations, blocking unwanted antibody binding. 4. Excluding dead cells. 5. Panel design, getting started, available tools.
